The arbovirus laboratory investigates the molecular epidemiology and ecology of arthropod-borne (arbo) viruses with projects in three areas:
Surveillance and control of arboviruses in Rio de Janeiro in preparation for the 2016 Summer Olympics (2013 - 2016)
Vector range, genetic traits and phylogeography of arboviruses in Danube Delta (2014 - 2020)
Human and animal diseases caused by arthropod-borne viruses (arboviruses) are of growing importance in many countries of Europe including Germany. Continuing eco-climatic changes and globalization create suitable conditions for the emergence of arboviruses in Germany. Thus, we initiated a program that compiles and analyses arbovirus and vector data collected over a number of successive years. This provided a solid base to determine the underlying causes of the seasonal fluctuations in arbovirus activity and the relative abundance of the mosquito vector species. This information can be used as a basis for vector control programs and might provide an early warning of the presence of arboviruses in Germany.
Since 2009, arbovirus surveillance was performed mostly in south-west Germany. So far, more than 350.000 mosquitoes were captured and assayed for the presence of arboviruses.
In 2009, Sindbis virus (SINV) was isolated from Culex and Anopheles mosquitoes that were exclusively trapped in the city of Weinheim, south-west Germany. SINV is the causative agent of a febrile illness in humans associated with maculopapular rash and joint pain. Consequently, a study was initiated to investigate the medical importance of SINV in that area. Only four out of 3389 investigated blood donor samples were tested positive for SINV-specific-IgG antibodies and all samples from 355 patients with clinically suspected acute SINV infections were tested negative for SINV-specific antibodies or SINV RNA, thus demonstrating the low medical importance of SINV in south-west Germany.
In 2009, Batai virus (BATV) was isolated from Anopheles maculipennis mosquitoes trapped around the village of Waghäusel. BATV may cause a mild illness among sheep and cattle. Thus, 195 serum samples from cattles around the village of Waghäusel were investigated for BATV-specific-IgG antibodies and 3 samples were tested positive, demonstrating past BATV infections.
In 2010, Usutu virus (USUV) was isolated from Culex pipiens pipiens mosquitoes trapped in the city of Weinheim. Since June 2011, considerable mortality in wild and captive bird species was observed in south-west Germany. Consequently, more than 400 dead birds were tested for the presence of USUV and USUV RNA was detected in more than 100 individuals from 6 species. Therefore, the mortality of birds was shown to be associated with the emergence of USUV. Consequently, a study was initiated to investigate the medical importance of USUV in that area. One out of 4200 investigated blood donor samples were tested positive for USUV-specific-IgG and -IgM antibodies. The early discovery of USUV in mosquitoes followed by the recent epidemic proof the importance and predictive value of our mosquito based monitoring program for zoonotic arboviruses.
In 2013 Germany launched a new program to implement longterm biosecurity projects under Global Partnership auspices: German Partnership Program for Excellence in Biological and Health Security
The Program is intended to help partner countries tackle biosecurity threats (e.g. bioterrorism, outbreaks of highly pathogenic diseases, pandemics). The aim is not only to prevent any misuse of infectious agents that could pose a threat not merely to Germany but also to strengthen the health services of our partner countries in Africa, Central Asia and South America, thus enhancing their national security.
Under this Program a range of projects are being implemented In Brazil in cooperation with various specialised Brazilian institutes (Federal University of Rio de Janeiro, Laboratório Central de Saúde Pública do Rio de Janeiro Noel Nutels, Oswaldo Cruz Foundation). Priority is given to six areas: awareness-raising; biosecurity; disease monitoring; detection and diagnostics; networking; capacity-building. With a budget of € 400.000 over four years (2013 - 2016), we are helping Brazil to set up an early warning system of dangerous and highly infectious diseases (dengue fever, Zika fever, chikungunya fever), stop their spread and rapidly identify and eliminate the vectors responsible.
The Danube Delta is the largest continuous marshland and the second largest and best preserved river delta in Europe. The Danube Delta also represents the place with the highest biodiversity and largest wetland in Europe and provides perfect environmental conditions necessary for a diverse and abundant mosquitoes fauna. Being located along major migratory bird routes, the Danube Delta represents an extremely important nesting and feeding zone for a large number of resident and migratory birds from Africa and Asia.
Mosquito-borne diseases are characterized as being mostly local and focal with transmission foci and hotspots which are influenced by the presence of the aquatic biotopes suitable for the development of mosquito populations in the areas where blood feeding occurs. Wild birds are important to public health, because they can act as biological or mechanical carriers of emerging zoonotic pathogens, either as a reservoir host or by dispersing infected arthropod vectors. Bird migration provides the necessary factors for the establishment of new endemic foci of disease (maintained through a local bird–arthropod life cycle) even at great distances from where the pathogen was acquired.
Given that the introduction and spread of the mosquito-borne viruses into a new geographic areas depends on the presence of infected hosts, suitable mosquito vector and favorable habitats, the knowledge of the resident mosquito species population, distribution, dynamics, their feeding preferences and vector competence are crucial to understand the epidemiological cycle and is useful in helping to predict future outbreaks of infections due to emerging zoonotic pathogens. In addition, this longitudinal mosquito surveillance project will help us to learn more about mosquito-borne pathogens ecology and their genetic and phylogeographic characterization.
