We investigate how arboviruses interact with their mosquito vectors and how these interactions affect virus evolution and transmissibility. We are studying the biotic and abiotic factors of arbovirus emergence and are involved in arbovirus outbreak response. In addition, we provide specific support for outbreak response to the WHO Collaborating Centre for Arbovirus and Haemorrhagic Fever Reference and Research including on-the-spot investigations, confirmation of diagnosis, handling of dangerous (biosafety level III) pathogens, case detection, and provision of logistics in the form of staff and supplies.

Research Projects

1. The German arbovirus surveillance and mosquito monitoring program (CULIMO and CULIFO)

Human and animal diseases caused by arthropod-borne viruses (arboviruses) are of growing importance in many countries of Europe including Germany (Schmidt-Chanasit et al 2012). Continuing eco-climatic changes and globalization create suitable conditions for the emergence of arboviruses in Germany (Becker et al 2015). Thus, we initiated a program that compiles and analyses arbovirus and vector data collected over a number of successive years. This provided a solid base to determine the underlying causes of the seasonal fluctuations in arbovirus activity and arbovirus emergence (Jöst et al. 2010 and 2011, Becker et al. 2012, Garigliany et al. 2015, Engel et al. 2016, Cadar et al. 2017, Lühken et al. 2017, Heitmann et al. 2018, Ziegler et al., 2019). In addition, the fundamental understanding of the mosquito vector ecology (Börstler et al 2016, Lühken et al. 2016, Leggewie et al. 2018) is the sine qua non to develop and implement sustainable vector and arbovirus control programs in Germany.

2. Surveillance of emerging arboviruses in Rio de Janeiro, Brazil (German Partnership Program for Excellence in Biological and Health Security)

In 2013 Germany launched a new program to implement longterm biosecurity projects under Global Partnership auspices: German Partnership Program for Excellence in Biological and Health Security. The program was intended to help partner countries tackle biosecurity threats (e.g. bioterrorism, outbreaks of highly pathogenic diseases, pandemics). The aim was not only to prevent any misuse of infectious agents that could pose a threat not merely to Germany but also to strengthen the health services of Brazil, thus enhancing Brazil´s national security. Under this Program a range of projects were implemented. In cooperation with various specialised Brazilian institutes (Federal University of Rio de Janeiro, Laboratório Central de Saúde Pública do Rio de Janeiro Noel Nutels, Oswaldo Cruz Foundation) we established an early warning system for arbovirus emergence that led to the early discovery of Zika virus in Rio de Janeiro and the better understanding of dengue virus, chikungunya virus and Zika virus ecology (Campos et al. 2016 and 2017, Heitmann et al. 2017), laboratory diagnostics (Huzly et al. 2016, Müller et al. 2016, Steinhagen et al. 2016, Corman et al. 2016, Müller et al. 2017, Rönnberg et al. 2017, Rockstroh et al. 2017, Rockstroh et al. 2018, Hansen et al. 2019, Rackow et al. 2019, Kareinen et al. 2019) and pathogenesis (Tappe et al. 2016, Gabriel et al. 2017, Röcker et al. 2018, Stanelle-Bertram et al. 2018, Cirne-Santos et al. 2019). Thus, we implemented early warning systems for arbovirus emergence in Rio de Janeiro (Ayllon et al. 2017), reduced arbovirus spread and rapidly identified and eliminated the vectors responsible.

3. Vector range, genetic traits and phylogeography of arboviruses in Danube Delta, Romania

The Danube Delta is the largest continuous marshland and the second largest and best preserved river delta in Europe. The Danube Delta also represents the place with the highest biodiversity and largest wetland in Europe and provides perfect environmental conditions necessary for a diverse and abundant mosquitoes fauna. Being located along major migratory bird routes, the Danube Delta represents an extremely important nesting and feeding zone for a large number of resident and migratory birds from Africa and Asia. Arboviruses are characterized as being mostly local and focal with transmission foci and hotspots which are influenced by the presence of the aquatic biotopes suitable for the development of arthropod populations in the areas where blood feeding occurs. Wild birds are important to public health, because they can act as biological or mechanical carriers of emerging arboviruses, either as a reservoir host or by dispersing infected arthropod vectors. Given that the introduction and spread of the arboviruses into new geographic areas depends on the presence of infected hosts, suitable arthropod vector and favorable habitats, the knowledge of the resident arthropod species population (Török et al 2016), distribution, dynamics, their feeding preferences and vector competence was studied to understand the enzootic cycles (Tomazatos et al. 2018). In addition, this longitudinal arthropod surveillance project helped us to learn more about abovirus ecology and their genetic and phylogeographic characterization.

4. Metagenomics profile and evolution of the viral communities in field-collected sand flies, mosquitoes and ticks from different remote ecosystems of the Amazonian rainforest, French Guiana

Different arthropod species (sand flies, mosquitoes and ticks) are vectors of a wide array of arboviruses (arthropod-borne viruses) and have likely been central to viral evolution. To better understand the extent of arthropod-borne pathogens as well as their origin and evolutionary history, it is crucial to uncover the full range of microbial agents, including viruses associated with arthropods. Our current knowledge of the diversity of sand flies-, mosquito- and tick-associated viruses is very scarce, in part due to the lack of investigation of arthropod viromes, at least in remote areas. The majority of the arboviruses sampled to date predominantly has a relationship with vertebrates or are known to be agents of diseases. Recent studies of arthropod RNA viruses have revealed that changes in genome size, structure and segmentation have occurred more frequently and on a larger scale than previously thought, with some arthropod viruses likely to be ancestors of those that infect vertebrates. Anthropogenic and natural changes in the environment and climate changes may in future bring humans closer to microorganisms they have not been previously in contact. The vast majority of viruses are thought to be undiscovered, and recent studies have shown that viruses discovered in insects are very divergent from known pathogens. This project will apply beside the entomological and ecological investigations a so called vector-enabled metagenomics approach on diverse arthropod vector species from different ecosystems of the Amazonian rainforest in French Guiana. It will also provide an unprecedented view of the virome diversity in the insect vectors from different remote and closed ecological habitats as well as its seasonal variability. The characterization of the novel viruses identified by high-throughput sequencing (Fernandez-Correa et al., 2019) will improve our knowledge on the molecular mechanisms of virus-host interactions in arthropod vectors, and will bring a new perspective on fundamental questions of virus origins, diversity, host range, genome evolution, and disease emergence.

5. Virome of ticks collected from domestic animals in districts of the Tigray Province, Ethiopia

Like in many developing countries, domestic animals play a crucial role in Ethiopia. They provide food in the form of meat and milk. Cattle are a source of cash income through sales of milk, meat and other related products. Furthermore, they act as storage of wealth and determine social status within the community. Despite the large number of cattle in the country, the sector is characterized by low productivity and, hence, income derived from this sector could not impart significant role in the development of Ethiopia´s economy. The low productivity is attributed to high disease incidence and parasite burden. Vector-borne diseases are major constraints to the development of viable livestock industries wherever they occur. Among these, tick and tick-borne diseases are widely distributed in Ethiopia, causing a tremendous economic importance in livestock production. Arboviruses could be responsible for diseases with serious health implications, welfare and socio-economic impact. The present study will focus on the known and novel arboviruses and their genetic characterization from ticks of cattle from Ethiopia. This will help to provide useful information to the concerned stake holders and to develop appropriate and sustainable control and prevention methods.

6. Sandfly-borne viruses from Israel and their impact on human and animal health (German-Israeli Foundation for Scientific Research and Development)

The general aim of this collaborative effort is focused on the characterization of Phlebotomine sandfly- borne viruses that infect humans in Israel and elucidating their medical importance. Specific objectives will be: (i) Set up country wide surveillance for Phlebotomus and Sergentomyia sandflies fitted to virus detection and blood meal analysis; (ii) Perform molecular and phylogenetic analysis of novel viruses identified in Phlebotomine sandflies in Israel and (iii) characterize the functional and clinical relevance of the identified sandfly-borne viruses.

7. Domestication of Aedes aegypti and the effect on virus diversity and distribution based on a field study in Benin, West Africa (DFG Africa Program)

As there are difficulties in pointing out specific genes that lead to increased vector competence, we conceived that vector competence is more likely the result of the effects of genetic adaptations for other functions being not linked directly with vector competence. Therefore, we will gain new insights in how the adaption to the human environment (for example host-feeding preference, oviposition or resting behavior) influences vector behavior and if there are consequences on virus prevalence. This will be accomplished within a field study in Benin. Ae. aegypti will be trapped along a transect reaching from urban sites with high anthropogenic impact, over rural sites with medium anthrophogenic impact to sylvatic sites with low anthropogenic impact. Changes in the basic transmission parameters of mosquitoes (e.g. host-feeding preference) associated with domestication are particularly important but poorly understood. The link between habitat change and adaptions of the vector and associated pathogens are essential to understand the emergence of viruses transmitted between humans and wildlife.
It was demonstrated that there are differences of virus prevalence in mosquitoes between disturbed and
natural habitats. There should also be a focus on viruses associated exclusively with mosquitoes, named mosquito-only viruses, which are unable to replicate in vertebrates and do not represent a health risk for animals and humans. Therefore, we do not focus only on known viruses, which are often described for West Africa (e.g. DENV or YFV). The emergence of ZIKV in Oceania and the Americas demonstrated the importance of an unbiased virus surveillance. Therefore, the study focus on the detection of known and unknown viruses (moboviruses and mosquito-only viruses) silently circulating in sylvatic and urban Ae. aegypti from Benin and their prevalence change according to the degree of domestication of
the vector. We postulate that the adaptations accompanying domestication have side effects that result in a varied prevalence of viruses in Ae. aegypti.

8. Establishment of an “Early Warning System for Mosquito-borne Diseases” and implementation of new vector control tools in the urban environment of Greece and Germany (EWSMD)

In this project, we will use the entomological, epidemiological, climatological and geographical data from Germany and Greece already available and/or to be produced by our partners, key actors in the field of pathogen detection and vector control in Europe, in order to develop a common Early Warning System for mosquito borne diseases to be implemented and evaluated in both countries. Substantial exchange of know-how and expertise will take place between two major research institutes in the field arbovirology and transfer to two Major mosquito control operators in Europe for operational purposes.
The project's objectives are: 1. Connecting major European actors in arbovirology and mosquito surveillance and control from Germany and Greece 2. Reanalyze the epidemiological data from the severe WNV epidemic in Northern Greece since 2010 and the USUV outbreaks in Germany since 2011 so as to prepare accordingly Germany and Greece against WNV, USUV and other arboviruses. 3. Identify high risk areas and critical time intervals for WNV and USUV circulation following modeling from entomological data and epidemiological data already produced by the partners during the last years and to be further developed for the years 2018 and 2019 (Lühken et al. 2017). 4. Provide good practices, innovative tools and methods for the control of container-breeding mosquitoes in urban settings, with special consideration of Culex pipiens and Aedes albopictus, which are the major European vectors for existing (USUV in Germany and WNV in Greece) and/or threatening (WNV for Germany, dengue, Chikungunya and Zika for both countries) mosquito-borne diseases. 5. Produce, update and make available this information to concerned Public Health Institutions and/or other bodies of both countries, a functional webGIS, in which the users will have the possibility to view, explore, query, and display the periodical risk maps of the Early Warning System for mosquito-borne diseases.

9. Vector competence of indigenous mosquitoes from Germany (German Environment Agency)

Within the framework of climate impact research, the vector competence of selected indigenous mosquito species for Sindbis, Usutu, Batai, West Nile, chikungunya, Zika,  and dengue viruses will be determined (Leggewie et al., 2016, Wagner et al., 2018). Thus,  as part of the laboratory work in the BSL-3 insectary mosquito infection and transmission rates for several arboviruses will be determined (Heitmann et al., 2018, Jansen et al., 2019). In addition, in vitro experiments will focus on the adaption of arboviruses to different temperatures. Based on the data obtained, a risk Analysis will be carried out and necessary preventive measures developed.
